Derbyshire lost three batsmen early, before du Plooy and Matt Critchley (45) combined to put on 93 for the fourth-wicket. Once the latter was run-out immediately after lunch, a flurry of wickets followed to leave du Plooy on 61 with Derbyshire 159-9.
A vital tenth-wicket partnership alongside Sam Conners (5 not out off 55 deliveries) saw du Plooy strike Jake Ball down the ground for six, while playing the ramp shot to Tom Barber for boundaries over the wicketkeeper.
It took a good catch from Joey Evison to remove du Plooy on 130, Derbyshire all out for 239. Conners claimed all three Nottinghamshire second innings wickets before the close, the hosts ending the day on 84-3, leading by 169.
