Billy Godleman hit an unbeaten 249 before Harry White took 4-40 to help put the Second XI in a commanding position ahead of the final day of the Championship match versus Nottinghamshire at The 3aaa County Ground.
After making 445 in their first innings, Derbyshire bowled their local rivals out for 132 with White and Greg Cork (3-22) particularly impressive.
Derbyshire chose to bat again as they reached 132-6 with Harvey Hosein hitting a half-century, before setting Notts 446 to win, with the visitors reaching 33-1 at the close.
Having resumed on 380-8, Godleman and Tom Taylor added a further 46 runs on the morning of Day Two before the latter was finally dismissed by Steven Mullaney for 76, bringing to an end a partnership of 240 which is the highest ninth-wicket partnership in the history of the Second XI Championship.
Harry White was the last man to go as Godleman finished unbeaten on 249 from 361 balls, hitting 27 fours and a six to help his side recover from 111-7 to 445 all out.
Derbyshire used just four seamers as they took to the field and they blew Notts away in 45 overs, bowling them out for just 132.
Billy Root, brother of Yorkshire and England batsman Joe Root, was the only player to get to grips with some good Derbyshire bowling as he fell one run short of his half-century.
White was too good for the three more senior members of the Notts batting lineup as he saw off Greg Smith, Mullaney and Will Gidman. Jonathan Clare (2-29) and Tom Taylor (1-31) were also in the wickets.
Derbyshire decided to bat again for a short period of time before declaring on 132-6 from 24 overs with Hosein unbeaten on 52, ably backed up by useful cameos from Tom Knight (27) and Godleman (20).
Notts were chasing a mammoth 446 to win the game, and they lost a wicket before the close as Taylor had Smith caught by Matt Critchley for 8. The visitors ended the day on 33-1, requiring a further 413 runs for victory.
