Tony Palladino took 6-29 to put Derbyshire in control as 21 wickets fell on Day Two of the Specsavers County Championship match at The 3aaa County Ground.
Palladino claimed his 14th five-wicket haul for the county as the hosts produced a dominant bowling display to dismiss Glamorgan for just 121 shortly after lunch.
Alex Hughes hit a well-made 57 – only the second half-century of the match – in reply, while Harvey Hosein scored 31 as Derbyshire posted 171 in their second innings, setting Glamorgan 302 to win.
Palladino then returned to remove Stephen Cook (5) before stumps to leave the visitors 16-1.
Glamorgan openers Connor Brown and Stephen Cook started the day on 20-0, but they were only able to add a further nine runs before Lockie Ferguson got the breakthrough. A trademark fast yorker from the New Zealand international castled Brown (14), before his fellow opener was dismissed two overs later.
Cook (9) edged a swinging delivery from Ravi Rampaul to Gary Wilson at second slip as the visitors slipped to 43-2.
Tom Cullen and Kiran Carlson then came together at the crease as they set about rebuilding the Glamorgan innings. The pair reached a 40-run partnership before Palladino came into the attack to remove Carlson for 23 as five wickets fell for 11 runs before lunch.
Palladino had his second shortly after as Cullen (20) was victim to a full and swinging delivery that was caught behind. Hughes then had Chris Cooke (1) out lbw, before Palladino dismissed both Graham Wagg and Ruaidhri Smith without scoring to reduce the visitors to 94-7 at the interval.
The fast bowler continued his fine form after the break to claim his second five-wicket haul in as many matches, dismissing Tim van der Gugten (9) and then David Lloyd (25) in consecutive overs. Palladino finished with his second best figures ever for Derbyshire and the first time he had claimed a five-wicket haul in successive first-class matches.
Kieran Bull was the last wicket to fall, without scoring, as Ferguson claimed his second of the match to ensure a 130-run first innings lead for Derbyshire.
However, Glamorgan started well in Derbyshire’s reply as Godleman (0) was out caught behind off van der Gugten in the third over.
Wayne Madsen was then out bowled for 14, before Tom Lace (24) fell on the stroke of tea to Lloyd as the hosts slipped to 64-3.
Wilson and Hughes’ partnership was worth 34 runs before Bull came into the attack to take two wickets in consecutive overs. Wilson (15) was caught by Hogan, before Critchley fell without scoring as he was caught by Brown at point.
Hughes continued his fine form to pass 50 for the fourth time this campaign, with his half-century coming from 79 balls, including eight boundaries.
Andersson (0) and Palladino (1) then both fell in successive overs as Bull claimed his fourth wicket of the match.
Harvey Hosein played a spirited innings to reach 31 before Michael Hogan returned to the attack to dismiss him and Rampaul without scoring in the last over as Derbyshire were 171 all out, setting Glamorgan a target of 302 runs to win.
After taking six in the first innings, Palladino opened the bowling for the hosts and he claimed another wicket in the final over of the day to leave the visitors requiring a further 286 runs to win.
