Hughes had already taken a smart catch off the bowling of Fynn Hudson-Prentice for the opening wicket of the match before the all-rounder dismissed James Bracey for eight.
Matt Critchley then claimed a brilliant caught and bowled to see off Jack Taylor, while Rampaul picked up his 22nd wicket of the competition by trapping Andrew Tye lbw in the final over.
Wayne Madsen (47) top-scored for Derbyshire, as they chased down 137 with 17 balls to spare to book their maiden place at Finals Day.
